The over- or under-winding of a DNA strand is referred to as DNA and is an indication of the strain on that strand. DNA supercoils before the replication fork as a result of the helicase's action during the unwinding of the helix during DNA replication. The replication fork progresses as this supercoiling grows. The supercoiling will actually stop the helicase from moving if it is not alleviated. The enzymes known as topoisomerases are essential for DNA replication and transcription because they regulate DNA topology. They take part in the DNA's over- or under-winding.
